Summary

In this study, Wong Su et al. used scanning ion conductance microscopy (SICM) to study real-time effects of aggregated alpha-synuclein (a-Syn) on neuroblastoma cell membranes. SICM imaging, performed with a Park NX12 AFM, revealed pore-like defects and morphological disruptions caused by both oligomeric and fibrillar a-Syn. Complementary AFM (non-contact, tapping mode) confirmed aggregate size and structure. SICM enabled non-invasive, nanometer-resolution visualization of dynamic membrane damage. This method provides a direct, label-free approach to studying a-Syn cytotoxicity, relevant to Parkinson’s disease pathology, and could extend to research in other neurodegenerative disorders.
